If my file’s URL is ‘mysite.com/wp-content/uploades/2018/05/importantfile.zip’ and I have created a password protected page using PHP dispatching will that protect my file’s URL from showing up in google searches?
Yes (as long as you are not linking to that file from some other place of your site)
It is a good idea to add an empty “index.html” page in that fold (where you are keeping the file).
If you don’t know what an empty index file is, you can download the following one from our site and use it (right click on the link and pick save as):

Also, if you are going to have many password protected files then it is a good idea to create a special folder in your site. Then put all those password protected files in that folder. Now you can put a .htaccess file in that folder with the following content so nothing in that folder is indexed:
Options -Indexes
